for Immune Derepression in Advanced Cancer<\/strong><\/span><\/h4>\n<div id=\"post-content-metadata\">\n<div class=\"post-type rightalign\"><\/div>\n<\/div>\n<p>\u00a0<span>Pterostilbene, being a methyl ether of resveratrol, is known to possess anti-inflammatory and anticancer activity in various model systems.<\/span><span>\u00a0 <\/span><span>It is known that in advanced cancer, excess inflammatory signaling may be associated with <\/span><g class=\"gr_ gr_22 gr-alert gr_gramm gr_inline_cards gr_disable_anim_appear Grammar only-ins replaceWithoutSep\" id=\"22\" data-gr-id=\"22\">reduction<\/g><span> in CD3 zeta chain signaling and inhibited <\/span><g class=\"gr_ gr_20 gr-alert gr_gramm gr_inline_cards gr_disable_anim_appear Grammar only-ins replaceWithoutSep\" id=\"20\" data-gr-id=\"20\">function<\/g><span> of natural killer (NK) cells.<\/span><\/p>\n<p>Given the importance of NK cells in the activity of various Immunotherapeutics, we sought to determine whether administration of a nanoparticle formulation of pterostilbene may reverse cancer associated suppression of NK activity.<span>\u00a0 <\/span>An initial study in <g class=\"gr_ gr_18 gr-alert gr_spell gr_inline_cards gr_disable_anim_appear ContextualSpelling ins-del\" id=\"18\" data-gr-id=\"18\">heathy<\/g> volunteers was performed to elucidate <g class=\"gr_ gr_19 gr-alert gr_gramm gr_inline_cards gr_disable_anim_appear Grammar only-ins replaceWithoutSep\" id=\"19\" data-gr-id=\"19\">amount<\/g> of NanoStilbene needed to be administered to achieve sufficient plasma concentration for induction of anti-inflammatory activity.<\/p>\n<p>Subsequent to this, the selected NanoStilbene dose was administered to twelve patients with advanced solid cancers for 3 weeks.<span>\u00a0 <\/span>Daily treatment with 300mgl of NanoStilbene caused <g class=\"gr_ gr_21 gr-alert gr_gramm gr_inline_cards gr_disable_anim_appear Grammar only-ins replaceWithoutSep\" id=\"21\" data-gr-id=\"21\">reduction<\/g> in serum levels of inflammatory markers TNF-alpha, IL-6, and CRP.<span>\u00a0 <\/span>Assessment of peripheral blood mononuclear cell ability to generate IFN-gamma subsequent to stimulation with anti-CD3 and anti-CD28 was increased.<\/p>\n<ul><\/ul>\n<\/div>\n<\/div>\n<\/div>\n<\/div>\n<div class=\"et_pb_row et_pb_row_1\">\n<div class=\"et_pb_column et_pb_column_1_2 et_pb_column_1    et_pb_css_mix_blend_mode_passthrough\">\n<div class=\"et_pb_module et_pb_text et_pb_text_1 et_pb_bg_layout_light  et_pb_text_align_left\">\n<div class=\"et_pb_text_inner\">\n<p><strong><span>12 patients with advanced cancer<\/span><\/strong><\/p>\n<ul>\n<li>ECOG &gt;2<\/li>\n<li>300mg NanoStilbene Oral daily (300mg PTER)<\/li>\n<li>Assessments pre and 1, 2, and 3 weeks after treatment<\/li>\n<\/ul>\n<p><strong><span>Inflammatory Markers Decreased<\/span><\/strong><\/p>\n<ul>\n<li>TNF, IL-6, CRP<\/li>\n<\/ul>\n<p><strong><span>Immune Markers Increased<\/span><\/strong><\/p>\n<ul>\n<li>IFN gamma from stimulated PBMC<\/li>\n<li>NK Cytolysis activity<\/li>\n<\/ul>\n<p>&nbsp;<\/p>\n<\/div>\n<\/div>\n<\/div>\n<div class=\"et_pb_column et_pb_column_1_2 et_pb_column_2    et_pb_css_mix_blend_mode_passthrough\">\n<div class=\"et_pb_module et_pb_image et_pb_image_0 et_always_center_on_mobile\"><span class=\"et_pb_image_wrap \"><img src=\"https:\/\/areyoucovidmune.com\/covidmune\/wp-content\/uploads\/2019\/12\/patients-1080x655-1.jpg\" alt=\"\" \/><\/span><\/div>\n<\/div>\n<\/div>\n<div class=\"et_pb_row et_pb_row_2\">\n<div class=\"et_pb_column et_pb_column_4_4 et_pb_column_3    et_pb_css_mix_blend_mode_passthrough et-last-child\">\n<div class=\"et_pb_module et_pb_text et_pb_text_2 et_pb_bg_layout_light  et_pb_text_align_left\">\n<div class=\"et_pb_text_inner\">\n<p><strong>TNF-a what is it?<\/strong><br \/><strong>Tumour Necrosis Factor alpha<\/strong> (<strong>TNF alpha<\/strong>), is an inflammatory cytokine produced by macrophages\/monocytes during acute inflammation and is responsible for a diverse range of signaling events within cells, leading to necrosis or apoptosis. The protein is also important for resistance to infection and cancers.<\/p>\n<p>TNF inhibitors are drugs that help stop inflammation. They\u2019re used to treat diseases like rheumatoid arthritis (RA), juvenile arthritis, psoriatic arthritis, plaque psoriasis, ankylosing spondylitis, ulcerative colitis (UC), and Crohn\u2019s disease. They\u2019re also called TNF blockers, biologic therapies, or anti-TNF drugs.<\/p>\n<\/div>\n<\/div>\n<div class=\"et_pb_module et_pb_image et_pb_image_1 et_always_center_on_mobile\"><span class=\"et_pb_image_wrap \"><img src=\"https:\/\/areyoucovidmune.com\/covidmune\/wp-content\/uploads\/2019\/12\/tnf-wk3-bct.jpg\" alt=\"\" \/><\/span><\/div>\n<div class=\"et_pb_module et_pb_text et_pb_text_3 et_pb_bg_layout_light  et_pb_text_align_left\">\n<div class=\"et_pb_text_inner\">\n<p><strong>IL-6 what is it?<\/strong><br \/><strong>IL-6<\/strong> or <strong>Interleukin 6<\/strong>, is an interleukin that acts as both a pro-inflammatory cytokine and an anti-inflammatory myokine. In humans, it is encoded by the IL6 gene. In addition, osteoblasts secrete IL-6 to stimulate osteoclast formation.<\/p>\n<p>Interleukin inhibitors are immunosuppressive agents which inhibit the action of interleukins. Interleukins are a group of cytokines which are synthesized by lymphocytes, monocytes, macrophages, and certain other cells. They function especially in the regulation of the immune system.<\/p>\n<\/div>\n<\/div>\n<div class=\"et_pb_module et_pb_image et_pb_image_2 et_always_center_on_mobile\"><span class=\"et_pb_image_wrap \"><img src=\"https:\/\/areyoucovidmune.com\/covidmune\/wp-content\/uploads\/2019\/12\/il6-wk3-bct.jpg\" alt=\"\" \/><\/span><\/div>\n<div class=\"et_pb_module et_pb_text et_pb_text_4 et_pb_bg_layout_light  et_pb_text_align_left\">\n<div class=\"et_pb_text_inner\">\n<p><strong>CRP what is it?<\/strong><br \/><strong>CRP<\/strong> or <strong>C-reactive Protein<\/strong> is an annular, pentameric protein found in blood plasma, whose circulating concentrations rise in response to inflammation. It is an acute-phase protein of hepatic origin that increases following interleukin-6 secretion by macrophages and T cells.<\/p>\n<p>Lowering of CRP may be seen with the use of statin drugs even without significant improvement of the cholesterol profile. NK cells play a major role in the host-rejection of both tumors and virally infected cells.<\/p>\n<p>NK cells are activated in response to interferons or macrophage-derived cytokines. They serve to contain viral infections while the adaptive immune response generates antigen-specific cytotoxic T cells that can clear the infection.
